I would upgrade to 5.00m33-6, it has some fixes in it plus a new memory stick driver that improves read times. Reformat the memory stick after you are finished with the upgrade yo insure readability with the stick.
Make sure that you have the UMD ISO MODE in the VSH menu set to either Sony NP9660 or the M33 driver, try both as sometimes one will work and the other will not. Also if need be try and put a UMD disk in the drive when trying to run it.
If possible check the ISO on another PSP to see if it does indeed work.
